SINGLE CELL MAPPING OF γδT CELLS AND VASCULATURE IN LYMPHOMA: A STUDY FOR LOCALIZATION AND FUNCTIONAL CHARACTERIZATION OF γδ LYMPHOCYTES INFILTRATING HUMAN LYMPHOMAS.

The vast majority of tumors are immunogenic and activate our immune system. The immuno-infiltration of tumors involves not only adaptive immunity mediated by αβ T lymphocytes but also innate immunity mediated by γδ T lymphocytes (gd TL) or NK cells. Tumor growth implicitly illustrates immuno-escape (IE) mechanisms.

For years, our team has paid particular attention to γδ TL lymphocytes. In this project, we make the main hypothesis that, in lymphoma, the biology of gd TL is disturbed, either qualitatively (reduced ability to recognize and/or destroy), or quantitatively (decreased infiltration and/or sequestration in areas distant from the tumor). Using innovative single cell “multiomics” approaches, this project will describe the distribution and functional status of all cytotoxic CD8+ T cells and  γδ TL effectors and NK cells in diffuse large cell B lymphomas.

The outcomes of this project will be to better understand the mechanisms by which the tumor escapes cytotoxic effectors, and thus define new immunotherapy approaches based on the amplification, recruitment and stimulation of TL.
